I.Nghiên cứu Protein Lúa mì Tầm quan trọng Nông nghiệp
Protein lúa mì là thành phần thiết yếu trong dinh dưỡng con người. Chúng là yếu tố quyết định chất lượng cuối cùng của các sản phẩm làm từ bột mì. Luận án này tập trung vào sự tích lũy của protein hạt lúa mì. Nghiên cứu khám phá cơ chế động học protein. Luân chuyển protein, tức là quá trình tổng hợp và phân hủy liên tục, đóng vai trò then chốt. Việc hiểu rõ những cơ chế này rất quan trọng. Nó giúp cải thiện hàm lượng và thành phần protein. Nông nghiệp hiện đại tìm kiếm các giống lúa mì năng suất cao. Đồng thời, chất lượng protein phải được đảm bảo. Mục tiêu là đáp ứng nhu cầu thị trường và người tiêu dùng. Luận án cung cấp dữ liệu khoa học chi tiết. Dữ liệu này hỗ trợ các chương trình nhân giống lúa mì. Việc này hướng tới việc tạo ra các giống mới với đặc tính protein tối ưu.
1.1. Mục tiêu Nghiên cứu Protein Lúa mì
Luận án xác định các yếu tố ảnh hưởng đến sự tích lũy protein. Mục tiêu là hiểu rõ sự đóng góp của luân chuyển protein. Nâng cao chất lượng dinh dưỡng của lúa mì là ưu tiên hàng đầu. Việc này cũng giúp cải thiện đặc tính công nghệ của bột mì. Phát triển các giống lúa mì tốt hơn là mục tiêu cuối cùng.
1.2. Đóng góp của Luận án Tiến sĩ
Nghiên cứu mang đến cái nhìn mới về quá trình tổng hợp và phân hủy protein. Nó cho thấy sự phức tạp của hệ thống protein trong hạt lúa mì. Các phát hiện có thể ứng dụng trong chọn tạo giống. Luận án cung cấp cơ sở dữ liệu quan trọng. Dữ liệu này hỗ trợ nghiên cứu sâu hơn về protein lúa mì.
1.3. Phương pháp tiếp cận khoa học

II.Tối ưu Hóa Hàm lượng Protein Lúa mì Vai trò Glutenin

Một phần nghiên cứu đã khảo sát việc đưa gen glutenin trọng lượng phân tử cao (HMW) vào. Việc này nhằm mục đích chọn lọc tái định hình proteome hạt lúa mì. Các glutenin HMW là thành phần chính của gluten. Chúng ảnh hưởng lớn đến độ bền và độ đàn hồi của bột mì. Phân tích proteomics đã được tiến hành. Nó giúp xác định những protein nào bị ảnh hưởng. Những thay đổi này có ý nghĩa quan trọng đối với chất lượng nướng bánh. Cấu trúc protein của glutenin tạo ra mạng lưới liên kết chéo. Mạng lưới này là nền tảng cho cấu trúc của bánh mì. 2.1. Tăng cường Glutenin trọng lượng phân tử cao

Việc đưa glutenin HMW không chỉ tăng lượng protein. Nó còn thay đổi toàn bộ hệ protein (proteome) của hạt. Phân tích sâu sắc đã xác định những protein nào bị điều hòa tăng hoặc giảm. Những thay đổi này có thể ảnh hưởng đến các đặc tính chức năng protein khác.
2.3. Ý nghĩa chất lượng bột mì
Sự thay đổi về thành phần glutenin trực tiếp cải thiện rheology bột mì. Bột có thể trở nên đàn hồi và bền hơn. Điều này rất có lợi cho quá trình nướng bánh. Chất lượng nướng bánh được nâng cao. Sản phẩm cuối cùng có thể có cấu trúc tốt hơn.
III.Hiểu biết Cấu trúc Protein Lúa mì Gluten và Thành phần
Gluten là phức hợp protein duy nhất có trong lúa mì. Nó chịu trách nhiệm về đặc tính viscoelastic (độ dẻo và độ đàn hồi) của bột mì. Gluten là yếu tố quyết định chất lượng nướng bánh. Cấu trúc protein của gluten bao gồm hai nhóm chính: gliadin và glutenin. Gliadin tạo độ dẻo và khả năng giãn nở. Glutenin mang lại độ bền và độ đàn hồi cho bột. Sự cân bằng và tương tác giữa gliadin và glutenin là rất quan trọng. Nó định hình rheology bột mì. Nghiên cứu sâu về cấu trúc protein của các thành phần này. 3.1. Gluten Thành phần cốt lõi của bột mì

3.3. Tương tác Protein và Rheology bột

Luân chuyển protein bao gồm tổng hợp và phân hủy protein. Quá trình này diễn ra liên tục trong suốt giai đoạn phát triển hạt. Nghiên cứu xác định tốc độ của hai quá trình này. Nó cho thấy sự biến động của từng loại protein theo thời gian. Hiểu được bản đồ luân chuyển này rất quan trọng. Nó cung cấp cái nhìn sâu sắc về cách protein tích lũy trong hạt. V.Ảnh hưởng Protein Lúa mì đến Chất lượng Nướng Bánh

Rheology bột mì, tức là các đặc tính lưu biến của bột, bị ảnh hưởng trực tiếp bởi chất lượng và số lượng protein. Các đặc tính chức năng protein như độ bền, độ đàn hồi và khả năng mở rộng của gluten rất quan trọng. Chúng ảnh hưởng đến từng bước trong quy trình làm bánh. Từ quá trình nhào trộn, ủ bột cho đến nướng. Nghiên cứu về protein lúa mì cung cấp kiến thức cần thiết. Kiến thức này giúp các nhà làm bánh và các nhà khoa học thực phẩm. Nó giúp họ tối ưu hóa công thức và quy trình sản xuất. Mục tiêu là đạt được sản phẩm nướng có chất lượng cao nhất.
5.1. Vai trò Gluten trong cấu trúc bánh

VI.Protein Lúa mì và Sức khỏe Bệnh Celiac Dị ứng Gluten
Protein lúa mì, đặc biệt là gluten, là nguồn gốc của một số vấn đề sức khỏe. Trong đó có bệnh Celiac và dị ứng gluten. Bệnh Celiac là một bệnh tự miễn nghiêm trọng. Nó gây tổn thương ruột non khi tiêu thụ gluten. Các gliadin là thành phần chính của gluten, thường được xác định là tác nhân gây bệnh. Dị ứng gluten là một phản ứng miễn dịch khác. Nó có thể gây ra nhiều triệu chứng. Chúng bao gồm khó tiêu, phát ban hoặc khó thở. Việc hiểu rõ cấu trúc protein của gluten và các thành phần của nó là rất quan trọng. 6.2. Nghiên cứu protein gây dị ứng
Gliadin, một phần của gluten, được xác định là tác nhân chính. Các nghiên cứu tập trung vào việc xác định các epitope gây độc. Việc này giúp phát triển các chiến lược làm giảm khả năng gây dị ứng của lúa mì. Phân tích cấu trúc protein là cần thiết.

Due to the negative correlation between wheat grain yield and grain protein content, low grain protein content has become a significant issue in some countries due to the breeding focus on the improvement of grain yield. It has been shown that the introgression of a high molecular weight glutenin subunit (HMW-GS) gene, Glu-1Ay, into commercial wheat cultivars can add 1% more protein to total grain content on a grain weight basis and improve bread-making quality without lowering grain yield.
Results within this thesis revealed the quantitative proteomic changes present in introgressed Glu-1Ay gene lines that underpin bread wheat protein content without changing other agronomic traits. It showed that the abundance of the 1Ay21* HMW-GS protein contributed less than 20% of the total protein increase observed. Of the 115 differentially expressed proteins (DEPs) found in Glu-1Ay lines, the majority are other storage proteins, and proteins involved in protein synthesis, protein folding, protein degradation, stress response and grain development. Gene co-expression analysis demonstrated that nearly half the DEP encoding genes strongly co-expressed during grain development.
Promoters of these genes are enriched in elements associated with transcription initiation and light response, such as CAAT-box, G- box, GATT-motif and the Gt1-motif. The final protein abundance in wheat grain comes from the relative rates of protein synthesis and protein degradation during grain development. To gain a more comprehensive and quantitative understanding on how different sets of proteins accumulate, a protein turnover atlas of wheat grain proteins during grain development was generated in this thesis by combining in vivo stable isotope labelling (15N) and in-depth quantitative proteomics. This atlas illustrated that although large variations of protein turnover rates were observed among individual proteins, they are tightly associated with protein abundance accumulation over time in different functional protein categories and different wheat grain tissues.
Functional category analysis showed that storage proteins are rapidly cycled, having both higher synthesis and degradation rates than other functional categories. By contrast, those proteins that participate in photosynthesis, DNA synthesis and glycolysis showed low synthesis and degradation rates at all times. Calculations from these rates show that 18% of total grain ATP production is used for grain proteome biogenesis and 2% for maintenance, and storage proteins alone consumed nearly half of these budgets. Due to the relatively high protein degradation rate of storage proteins as a class of grain proteins, it was calculated that 25% of newly synthesized storage proteins were degraded during grain development and thus were absent at harvest.
These VIII data show that stabilization of the wheat grain proteome could improve grain protein content without introducing extra protein synthesis costs. Overall, the findings obtained from this thesis provide a quantitative insight into which sets of wheat proteins accumulate in high protein genotypes and a new perspective on the energy use efficiency of protein synthesis and maintenance (PEUE). The data helps fill the knowledge gap of the spatiotemporal variation of wheat grain protein abundance that cannot be directly explained by transcript abundance and can also direct selection of storage protein candidates with higher PEUE for future wheat quality breeding programmes.
