I. Tương tác sắc tố mật Bảo vệ chống chất gây đột biến

Ung thư là một nguyên nhân hàng đầu gây tử vong ở người. Tiếp xúc với các hợp chất gây đột biến môi trường đóng vai trò quan trọng trong quá trình phát triển bệnh. Câu hỏi đặt ra là làm thế nào con người có thể tự bảo vệ khỏi các chất độc hại này. Gần đây, các sắc tố mật (BPs) như biliverdin, bilirubin không liên hợp và protoporphyrin đã được phát hiện. Chúng hoạt động như chất chống oxy hóa. Chúng ức chế tác động gây đột biến của một số chất gây đột biến môi trường nổi tiếng. Các chất này bao gồm 2-aminofluorene, benzo[α]pyrene và 2-amino-1-methyl-6-phenylimido[4,5-b]pyridine. Mặc dù kết quả đầy hứa hẹn, cơ chế ức chế vẫn chưa được biết rõ. 1.1. Mối đe dọa từ chất gây đột biến đa vòng

Tiếp xúc với các chất gây đột biến môi trường là một nguyên nhân chính gây ung thư ở người. Các Hydrocarbon thơm đa vòng (PAHs) là một nhóm chất gây đột biến đa vòng phổ biến. Chúng có khả năng gây độc tính di truyền cao. Benzo[α]pyrene là một ví dụ điển hình của PAHs. Sự hiện diện của các chất này trong môi trường đặt ra một thách thức lớn cho sức khỏe cộng đồng. Việc tìm kiếm các phương pháp bảo vệ hiệu quả là cần thiết để giảm thiểu rủi ro gây ung thư.

1.2. Vai trò sắc tố mật trong chống đột biến

Các sắc tố mật (Bilirubin, Biliverdin, Protoporphyrin) là các hợp chất tự nhiên trong cơ thể. Chúng được biết đến với đặc tính chống oxy hóa. Gần đây, khả năng ức chế hiệu quả gây đột biến của chúng đã được ghi nhận. Các sắc tố mật có thể chống lại tác động của nhiều chất gây đột biến đã biết. Điều này mở ra hướng nghiên cứu mới về tiềm năng của sắc tố mật trong việc phòng ngừa ung thư. Tuy nhiên, cơ chế chính xác đằng sau khả năng ức chế này vẫn còn là một bí ẩn. II. Đánh giá tương tác vật lý sắc tố mật và mutagens

Các phương pháp bao gồm cộng hưởng từ hạt nhân (NMR), quang phổ UV và đánh giá tác động lên khả năng sinh khả dụng. 2.1. Phân tích bằng phổ NMR và UV của sắc tố mật

Phổ NMR của chất gây đột biến đã được so sánh khi có và không có sắc tố mật. Rất ít thay đổi về dịch chuyển hóa học của các proton đã được ghi nhận. Những thay đổi nhỏ xảy ra là kết quả của tương tác acid/base giữa sắc tố mật và chất gây đột biến. Phổ UV của mỗi chất gây đột biến cũng được đo. Việc này được thực hiện khi có và không có các nồng độ sắc tố mật khác nhau. Không có thay đổi nào trong phổ UV của bất kỳ hợp chất nào được quan sát. Các kết quả này cho thấy không có tương tác vật lý mạnh hoặc sự kết tụ đáng kể giữa các phân tử.

2.2. Ảnh hưởng đến khả dụng sinh học chất gây đột biến

Tương tác vật lý mạnh hoặc sự kết tụ của các hợp chất có thể ảnh hưởng đến sự hấp thu của chúng qua các lớp tế bào đơn. Vì vậy, khả năng thấm biểu kiến của chất gây đột biến qua các lớp tế bào Caco-2 đã được đo. Việc này được thực hiện khi có và không có sắc tố mật. III. Cơ chế hóa học sắc tố mật loại bỏ chất gây đột biến

Các phản ứng hóa học giữa sắc tố mật và các chất chuyển hóa gây tổn thương DNA của chất gây đột biến đã được nghiên cứu. Styrene epoxide đã được sử dụng làm mô hình cho các chất chuyển hóa phản ứng. Styrene epoxide có sẵn trên thị trường, ổn định và ít độc hơn các chất chuyển hóa phản ứng của chất gây đột biến thực tế. Các phản ứng cạnh tranh đã được thực hiện. Sắc tố mật và các dẫn xuất của chúng được đặt trong dung dịch với guanine. Sau đó, chúng được cho phản ứng với styrene epoxide. Các phản ứng này đã chỉ ra rằng sắc tố mật và các ester dimethyl của chúng phản ứng với epoxide mạnh hơn guanine. Sắc tố mật chủ yếu phản ứng thông qua các nhóm axit cacboxylic của chúng. Mono- và di-styrene epoxide esters là các sản phẩm chính được phân lập từ các phản ứng. Các vòng pyrrole trong bilirubin cũng cho thấy một số bằng chứng về phản ứng với styrene epoxide. 3.1. Sắc tố mật và phản ứng với epoxide chất gây đột biến

Các sắc tố mật và các dẫn xuất của chúng đã được nghiên cứu về khả năng phản ứng hóa học với các chất chuyển hóa epoxide. Các thí nghiệm cạnh tranh đã được thực hiện. Sắc tố mật phản ứng mạnh hơn guanine với styrene epoxide. Điều này cho thấy khả năng của sắc tố mật trong việc loại bỏ các chất chuyển hóa gây tổn thương DNA. 3.2. Khả năng loại bỏ chất chuyển hóa phản ứng sắc tố mật

Sắc tố mật có khả năng loại bỏ các chất chuyển hóa phản ứng. Tuy nhiên, nhóm cacboxylic tự do cho thấy hiệu quả cao hơn đáng kể. Các dẫn xuất ester dimethyl không đạt được hiệu quả tương tự. Hơn nữa, sự phổ biến của các nhóm cacboxylic acid trong môi trường tế bào gợi ý rằng cơ chế loại bỏ hóa học này không phải là duy nhất. IV. Ức chế chuyển hóa chất gây đột biến bởi sắc tố mật

Một cơ chế hoạt động tiềm năng khác của sắc tố mật là ức chế sự hình thành các chất chuyển hóa gây tổn thương DNA của chất gây đột biến. Để kiểm tra giả thuyết này, các thí nghiệm in vitro đã được thực hiện. Chất gây đột biến được đồng ủ trong phân đoạn S9 gan người. Các thí nghiệm được thực hiện khi có và không có sắc tố mật. Kết quả đã chỉ ra rằng sắc tố mật là chất ức chế chuyển hóa của benzo[a]pyrene và 2-amino-1-methyl-6-phenylimido[4,5-b]pyridine. Sự ức chế này được thực hiện bởi các enzyme gan. Thứ tự hiệu quả ức chế là bilirubin > protoporphyrin > biliverdin. Các nghiên cứu mô hình phân tử cũng đã được thực hiện. Chúng kiểm tra sự gắn kết của các sắc tố mật khác nhau vào các vị trí hoạt động của các cấu trúc tinh thể enzyme đã công bố. Các enzyme này được biết là chịu trách nhiệm chuyển hóa chất gây đột biến. 4.1. Sắc tố mật ức chế chuyển hóa benzo a pyrene

Các sắc tố mật được chứng minh là chất ức chế quá trình chuyển hóa các Hydrocarbon thơm đa vòng. Cụ thể, chúng ức chế sự chuyển hóa của Benzo[a]pyrene và 2-amino-1-methyl-6-phenylimido[4,5-b]pyridine. Quá trình này được thực hiện bởi các enzyme trong gan người. Thứ tự hiệu quả ức chế của các sắc tố mật là Bilirubin > Protoporphyrin > Biliverdin. Điều này cho thấy Bilirubin có khả năng ức chế chuyển hóa chất gây đột biến đa vòng mạnh nhất. Việc ức chế này làm giảm sự hình thành các chất chuyển hóa phản ứng, từ đó giảm độc tính di truyền.

4.2. Nghiên cứu mô hình phân tử tương tác sắc tố mật

Các nghiên cứu mô hình phân tử đã được tiến hành để hiểu rõ hơn về Tương tác hóa học. Các sắc tố mật đã được mô phỏng để gắn vào các vị trí hoạt động của các enzyme chuyển hóa chính. Các enzyme này bao gồm CYP1A1, 1A2, 1B1 và 3A4. Kết quả cho thấy sắc tố mật có khả năng liên kết hiệu quả với các vị trí này. Điều này hỗ trợ giả thuyết rằng sắc tố mật ức chế hoạt động của enzyme. Việc ức chế enzyme này làm giảm đáng kể sự hình thành các chất chuyển hóa gây đột biến. Từ đó, chúng giảm nguy cơ gây ung thư.

V. Tóm tắt cơ chế chống đột biến của sắc tố mật

Nghiên cứu đã tiến hành một loạt các thí nghiệm để đánh giá các cơ chế có khả năng giải thích tác động ức chế của sắc tố mật đối với các chất gây đột biến môi trường đã biết. Các phát hiện cho thấy tương tác vật lý không đóng vai trò chính. Việc loại bỏ hóa học các chất chuyển hóa phản ứng cũng không phải là cơ chế độc quyền hoặc chủ yếu. Thay vào đó, sự ức chế chuyển hóa của chất gây đột biến bởi sắc tố mật là cơ chế quan trọng nhất. Điều này cho thấy sắc tố mật có tiềm năng lớn trong việc ngăn ngừa độc tính di truyền và quá trình gây ung thư bằng cách tác động trực tiếp lên quá trình trao đổi chất của các hợp chất gây hại. 5.1. Các cơ chế được đề xuất sắc tố mật

5.2. Kết luận về cơ chế chính của sắc tố mật

Physical and chemical interactions between bile pigments and polyaromatic mutagens Hung Trieu Hong BSc. and Master in Organic Chemistry A thesis submitted for the degree of Doctor of Philosophy at The University of Queensland in 2015 School of Chemistry and Molecular Biosciences Abstract A major cause of cancer in humans is exposure to mutagenic compounds. This raises the question of how humans can be protected from these environmental mutagens. Bile pigments (BPs) such as biliverdin, unconjugated bilirubin and protoporphyrin and their derivatives have recently been found to act as antioxidants and inhibit the mutagenic effects of several known environmental mutagens including 2-aminofluorene, benzo[α]pyrene, and 2-amino-1-methyl-6-phenylimido[4,5- b]pyridine.

Despite these promising results, very little is known about the mechanisms by which this inhibition is achieved. Understanding these mechanisms would be useful for future drug development. Therefore, this PhD thesis aims to explore physical and chemical interactions between BPs and mutagens. Effects of BPs on the bioavailability and metabolism of mutagens were also examined in vitro using the colorectal adenocarcinoma (Caco-2 cell) monolayer model and the human liver S9 fraction.

The physical interactions between mutagens and BPs were examined using three different methods: NMR, UV and effects of bioavailability. The results of the comparison of the NMR spectra of mutagens in the absence and presence of BPs showed very little changes in the chemical shifts of the protons and the changes that did occur were the result of acid/base interactions between the BPs and mutagens. The UV spectrum of each mutagen was measured in the presence and absence of varying concentrations of BPs, and there were no changes to the UV spectra of any of the compounds. Strong physical interactions or aggregation of compounds can also affect their absorption across cell monolayers and so the apparent permeability of mutagens across Caco-2 cell monolayers in the presence and absence of BPs were measured.

The results indicated that BPs increased the permeability of the mutagens slightly and effected how much of the compounds remained in tight association with the monolayer but the effects were small. These experiments provided evidence to suggest that physical interactions and aggregations are unlikely to be a major contributing mechanism of the inhibitory effects of BPs on environmental mutagens. Chemical reactions between BPs and the DNA modifying metabolites of mutagens (epoxides) were studied using styrene epoxide as a model for the reactive metabolites. Styrene epoxide is commercially available, stable and less toxic than the reactive metabolites of the mutagens.

Competitive reactions were performed in which BPs and their derivatives were placed in solution with guanine and allowed to react with styrene epoxide. These reactions showed that BPs and their dimethyl esters are more reactive to the epoxide than guanine. Bile pigments primarily react through their carboxylic acid groups with the mono- and di-styrene epoxide esters being the major products isolated form the reactions. The pyrrole rings in bilirubin also showed some evidence of ii reaction with styrene epoxide though the products were too unstable to isolate.

Thus, it was clear that BPs can effectively scavenge reactive metabolites, but the free carboxylic acids were significantly more effective at this than the dimethyl ester derivatives. This is not reflected in the anti-mutagenic activities of the compounds. Also, the ubiquitous nature of carboxylic acid groups in the cellular environment makes it unlikely that this reaction with activated epoxides would be unique to BPs. For these reasons we concluded that it is unlikely that chemical scavenging of reactive metabolites is the sole or even major mechanism of the inhibition of BPs.

Another possible mechanism of action of the BPs is that they inhibit the formation of the DNA modifying metabolites of the mutagens. We investigated this by performing in vitro experiments in which mutagens were co-incubated in the human liver S9 fraction in the presence and absence of BPs. The results indicated BPs were inhibitors of the metabolism of benzo[a]pyrene and 2-amino-1- methyl-6-phenylimido[4,5-b]pyridine by liver enzymes. The order of inhibitory effectiveness was bilirubin > protoporphyrin > biliverdin.

Molecular modelling studies which examined the docking of the various BPs into the active sites of published crystal structures of the enzymes known to be responsible for the metabolism of the mutagens, suggested BPs could bind to the active sites of CYP1A1, 1A2, 1B1 and 3A4. In summary, we conducted a series of experiments to evaluate the likely mechanisms of the inhibitory effects of BPs on known environmental mutagens. There are several theories postulated to explain the anti-mutagenic effects of BPs including the physical -stacking driven aggregation of BPs with the polyaromatic mutagens, the chemical scavenging of BPs towards reactive metabolites, and the inhibition of BPs of the P450 mediated activation of the mutagens. We have systematically tested each of these and found that the latter appears to be the most likely mechanism to explain the effects reported.

In broader terms, this research will aid in understanding how BPs inhibit mutagenesis and thus may lead to the development of synthetic compounds that could decrease the risk to humans exposed to these environmental mutagens. iii Declaration by author This thesis is composed of my original work, and contains no material previously published or written by another person except where due reference has been made in the text.
