I.Công nghệ quang học nano cho chẩn đoán sinh hóa

Công nghệ quang học nano đang cách mạng hóa lĩnh vực chẩn đoán sinh hóa. Các cảm biến sinh học quang học nano cung cấp khả năng phát hiện nhanh chóng, chính xác và nhạy bén. Chúng cho phép phân tích không nhãn, giảm thiểu các bước chuẩn bị mẫu và tăng tốc quá trình xét nghiệm sinh hóa. Việc ứng dụng các thiết bị quang học nano mang lại những nền tảng nhỏ gọn, mạnh mẽ cho nhiều ứng dụng khác nhau. Các đầu dò sinh học quang học đóng vai trò quan trọng trong việc theo dõi thời gian thực các tương tác sinh học. Sự phát triển này thúc đẩy việc tạo ra các công cụ chẩn đoán hiệu quả hơn và dễ tiếp cận hơn.

1.1. Ưu điểm nổi bật của cảm biến quang học

Công nghệ quang học mang lại nhiều lợi thế đáng kể trong chẩn đoán sinh hóa. Chúng cho phép phát hiện nhanh, chính xác và có độ nhạy cao. Lợi ích chính bao gồm hoạt động không nhãn. Điều này cho phép phân tích trực tiếp mà không cần thẻ huỳnh quang hoặc đồng vị phóng xạ. Quá trình này đơn giản hóa việc chuẩn bị mẫu và giảm độ phức tạp của xét nghiệm. Các cảm biến quang học rất dễ thích nghi để thu nhỏ kích thước. Điều này dẫn đến các thiết bị chẩn đoán nhỏ gọn và di động. Công nghệ này cung cấp khả năng giám sát thời gian thực, rất quan trọng cho các quá trình sinh học động. 1.2. Các phương pháp cảm biến quang học chủ đạo

Một số phương pháp cảm biến quang học nổi bật trong phân tích sinh hóa. Cộng hưởng plasmon bề mặt (SPR) là một kỹ thuật hàng đầu. Nó đo lường sự thay đổi chỉ số khúc xạ gần bề mặt kim loại. Phổ huỳnh quang phát hiện ánh sáng phát ra từ các phân tử bị kích thích, mang lại độ nhạy cao. Các thiết bị ống dẫn sóng dẫn ánh sáng qua một môi trường. Chúng cảm nhận các tương tác trên bề mặt của mình. Những phương pháp đa dạng này tạo nên nền tảng của ghép cặp cộng hưởng quang học nano. Chúng tạo điều kiện thuận lợi cho việc tạo ra các cảm biến sinh học hiệu quả cao. Sự tích hợp quang học nano tăng cường hiệu suất của các kỹ thuật này. Điều này dẫn đến các công cụ phân tích tiên tiến. Những công cụ này rất quan trọng cho việc phát triển các xét nghiệm sinh hóa thế hệ tiếp theo.

II.Cộng hưởng Plasmon Bề mặt SPR trong xét nghiệm

Cộng hưởng Plasmon Bề mặt (SPR) là một công nghệ cốt lõi trong các xét nghiệm sinh hóa hiện đại. Nó khai thác sự dao động tập thể của electron tại giao diện kim loại-điện môi. Đây là các plasmon bề mặt, rất nhạy cảm với những thay đổi trong môi trường xung quanh. Công nghệ plasmonics này cho phép phân tích sinh hóa không nhãn và phát hiện dấu ấn sinh học với độ nhạy cao. Sự kích thích SPR tạo ra một trường evanescent tương tác với các phân tử, cung cấp thông tin thời gian thực về các tương tác. Khả năng định lượng của SPR làm cho nó trở thành một công cụ không thể thiếu trong nghiên cứu và chẩn đoán.

2.1. Nền tảng và nguyên lý kích thích SPR

Cộng hưởng Plasmon Bề mặt (SPR) hình thành nên cốt lõi của nhiều cảm biến sinh học tiên tiến. SPR liên quan đến sự dao động tập thể của electron tại giao diện kim loại-điện môi. Những dao động này, được gọi là plasmon bề mặt, nhạy cảm với sự thay đổi chỉ số khúc xạ của môi trường lân cận. Khi ánh sáng tương tác với màng kim loại ở một góc và bước sóng cụ thể, cộng hưởng xảy ra, hấp thụ năng lượng ánh sáng tới. Điều kiện này được gọi là khớp pha. Cấu hình Kretschmann, sử dụng lăng kính, là một phương pháp phổ biến để kích thích SPR. Ánh sáng bị phản xạ toàn phần bên trong, tạo ra một trường evanescent kết nối với các plasmon bề mặt. Độ sụt giảm cường độ ánh sáng phản xạ cho thấy góc SPR. Hiện tượng này cung cấp một nền tảng có độ nhạy cao để phát hiện các sự kiện liên kết phân tử.

2.2. Hiệu suất và ứng dụng của cảm biến SPR

Các cảm biến SPR mang lại hiệu suất vượt trội cho phân tích sinh hóa. Chúng cho phép phát hiện tương tác phân tử theo thời gian thực, không nhãn. III.Nâng cao phát hiện huỳnh quang bằng cộng hưởng SPR

Việc nâng cao phát hiện huỳnh quang thông qua cộng hưởng plasmon bề mặt (SPR) là một tiến bộ quan trọng. Kỹ thuật này sử dụng trường evanescent của SPR để khuếch đại tín hiệu huỳnh quang, cải thiện đáng kể độ nhạy của các xét nghiệm. Nó rất hữu ích cho phát hiện dấu ấn sinh học ở nồng độ thấp. Các cảm biến nano quang được thiết kế với cấu hình cụ thể. Điều này tối ưu hóa sự tương tác giữa plasmon và chất huỳnh quang. Thiết bị quang học nano này cung cấp một nền tảng mạnh mẽ cho việc phát triển các đầu dò sinh học quang học tiên tiến. Điều này giúp đẩy nhanh các phân tích sinh hóa nhanh và chính xác.

3.1. Động lực và cơ chế tăng cường huỳnh quang

Việc tăng cường độ nhạy phát hiện huỳnh quang là một mục tiêu quan trọng trong các xét nghiệm sinh hóa. Huỳnh quang thông thường thường bị tín hiệu yếu, đặc biệt với các chất phân tích khan hiếm. IV.Cảm biến không nhãn Sợi quang cho phân tích sinh hóa

Cảm biến không nhãn dựa trên sợi quang đại diện cho một bước tiến trong phân tích sinh hóa. Các cảm biến nano quang này loại bỏ nhu cầu về các phân tử đánh dấu. Chúng hoạt động bằng cách cảm nhận trực tiếp những thay đổi trong chỉ số khúc xạ của môi trường xung quanh. Điều này xảy ra khi các phân tử sinh học liên kết với bề mặt sợi. Phương pháp này cung cấp các thiết bị quang học nano nhỏ gọn và có khả năng giám sát thời gian thực. Chúng là các đầu dò sinh học quang học lý tưởng cho các xét nghiệm sinh hóa nhanh. Công nghệ này đặc biệt hiệu quả trong việc phát hiện các tương tác protein, góp phần vào sự phát triển của công cụ chẩn đoán hiệu quả.

4.1. Khái niệm cảm biến không nhãn qua sợi quang

Các cảm biến sinh học không nhãn cách mạng hóa phân tích sinh hóa. Chúng loại bỏ nhu cầu về các thẻ phân tử. Các cảm biến dựa trên sợi quang cung cấp một nền tảng linh hoạt cho mục đích này. Chúng hoạt động bằng cách điều biến các tính chất quang học của lớp phủ sợi. Sự thay đổi chỉ số khúc xạ của môi trường xung quanh, do liên kết phân tử, ảnh hưởng trực tiếp đến sự truyền ánh sáng bên trong sợi. Sự điều biến này cho phép phát hiện tương tác sinh học phân tử theo thời gian thực mà không làm thay đổi trạng thái tự nhiên của chất phân tích. Các cảm biến như vậy nhỏ gọn, mạnh mẽ và tiết kiệm chi phí. Chúng lý tưởng cho cảm biến từ xa và chẩn đoán tại điểm chăm sóc. Trọng tâm là điều biến lớp phủ lỏng. Trong đó, chính dung dịch chất phân tích đóng vai trò là lớp phủ nhạy cảm, tương tác trực tiếp với trường evanescent.

4.2. Hệ thống và khả năng phát hiện protein BSA

Một hệ thống cảm biến quang học sợi không nhãn bao gồm các kỹ thuật biến đổi bề mặt cụ thể. Liên kết BSA đòi hỏi sự chuẩn bị cẩn thận bề mặt sợi. Vật liệu SiO2 polymer lõi, khi được sử dụng làm lớp phủ nhạy cảm, cho thấy các đặc tính hấp thụ cụ thể với dung dịch nước-glycerin. Thiết bị thực nghiệm sử dụng sợi đa mode. Trong đó, các tương tác giữa lõi sợi nhựa và môi trường thử nghiệm (protein BSA) được phân tích. Phân tích này tiết lộ độ nhạy của cảm biến. Khả năng của hệ thống phát hiện protein BSA mà không cần nhãn xác nhận tiềm năng của nó cho các ứng dụng sinh hóa rộng hơn. Các cảm biến sinh học sợi quang như vậy đóng góp đáng kể vào việc phát triển các công cụ chẩn đoán nhanh và chính xác. Điều này thúc đẩy ranh giới của ghép cặp cộng hưởng quang học nano.

We present two types of optical diagnostic devices with their performance, i., the label-free device and fluorescence detection (label-aided) based ones for quantitative assays of biomolecules including proteins and DNAs. First, we present the label-free optical biosensors based on optical fibers, which sensitively respond to introduction of analyte molecules for point-of-care-testing (POCT) application that demands the real-time quantitative diagnosis of biochemical analyte concentration in a compact device structure. We demonstrate the modulation of fiber optical modes by means of liquid cladding which is formed by introducing analyte liquid into the fiber core.

Unlike other types of label-free optical sensors such as optical fiber sensors based on surface plasmon resonance (SPR), the presented optical sensors do not need any additional coatings on its core, such as thin metal film or high index dielectric films, with the great advantages of x both the realization of the simple structure device and highly reproducible sensor performance. The fiber (waveguide) mode mismatch between interfaces of different claddings can be used for highly sensitive change in optical transmission, allowing the label- free bio-chemical diagnosis to be sensitive enough, making these sensors comparable to the plasmonic fiber sensors. It is also unveiled that the device presented has the great inherent advantages of the device-length independence of sensing performance in contrast to the other types of fiber based sensors. We describe its working principle by building up the mathematical modeling of waveguide mode mismatch and liquid analyte absorption of evanescent fields.

We experimentally achieve the limit of detection (LOD) of glycerol concentration of 0.02 × 10-6 refractive index unit) and LOD of BSA concentration of 3. This dissertation also reports the enhancement of fluorescence by near-field coupling of fluorophores with surface plasmons using two different fluorescence detection platforms, i., an upright and inverted types of a flurorescence microscopes. Surface plasmons that enhance the fluorescence and reduce the photobleaching, can be generated by nanoscale thick films of metal layers deposited on a xi slide glass used in a microscope. We investigate various metal thinckesses of gold, silver and their combination to optimize the decaying depth of the evanescent field SPR wave into the sensing region where fluorophores are dispersed.

It is found that the bimetallic layer of silver (52 nm) and gold (2 nm), which is determined by numerical calculation, provides the maximized depth of the SPR evanescent field with strongest coupling of fluorophores with the plasmons at both spectral channels of excitation (470 nm) and emission (525 nm) wavelengths. We experimentally demonstrate the enhancement of fluorescence emitting from interferon- double stranded DNAs (283 bp, polymerase chain reaction product) conjugated with the fluorophores (SYBR Green I), with the enhancement factor estimated as 45-fold. It is noted that this enhancement is achieved only by plasmonic coupling with fluorophores at the emission channel without excitation enhancement. This enhancement can lead to the LOD of DNAs concentration of ~80 fg/L (1.6 nM) in an upright format of a fluorescence miscroscope.

We also achieve the extended photobleaching-free range of DNA concentrations in the fluororescence based assays, allowing photostable diagnosis of the analytes. xii KEYWORDS: Optical biosensor; label-free sensor; fiber optical sensor; Nano-biophotonics; Surface Plasmon Resonance; fluorescence enhancement; fluorescence microscope, DNA assay. INTRODUCTION TO OPTICAL TECHNOLOGY FOR BIOCHEMICAL DIAGNOSIS 1. Surface plasmon resonance (SPR) Althouhg the phenomenon of SPR was known more than a century ago.

In 1902, Wood observed an uneven distribution of light in a reflected spectrum from a metallic diffraction grating [Wood, 1902]. The 60 years later, Thurbadar recognized the adsorption in the reflectivity of a metal film [Turbadar, 1959]. However, both of them did not identify these observations as surface plasmon resonance (SPR). The original of surface plasmon is based on the Maxwell’s equations, where the free electrons charge of the metal are considered as the plasma. Measurement of the electron-plasma oscillation in metals is highly dependent on the dimensions of aggregative longitudinal 1 excitations of the conductive free-electron gas (see Figure 1.1), and plasmon are the quanta of such charge-density oscillations over a surface. The charge-density fluctuations can be existed in the bound at the interface between metal and dielectric, where they propagate as waves along the interface forming what called surface plasmon.
